When it comes to practicing for your audition; there is no wrong or right way to approach it. Practice no matter what, because not practicing can hurt you more when it comes time to perform your part for the audition. You can even take and practice in front of a mirror with whatever script or monologue you have prepared. Then you can move on to practice your lines in front of other people; who can give you objective feedback and criticism about your performance. You could even film yourself and watch yourself read the script or monologue to gage your own strong and weak points that need to be corrected before the audition.
